COMPOSITION:
Penicillin V 500 : Each tablet contains : 500000 IU equivalent to 312.5mg
Penicillin V as potassium.
Penicillin V 1000 : Each tablet contains : 1000000 IU equivalent to 625mg
Penicillin V as potassium.
Penicillin V 400 : Each 5 ml contains: 400000 IU equivalent to 250 mg
Penicillin V as potassium.
PROPERTIES:
Penicillin V potassium is a beta-lactam antibiotic and has a bacterial action against Gram-positive bacteria. Gram-negative cocci, some other Gram-negative bacteria, spirochetes, and actinomycetes. It inhibits bacterial cell-wall synthesis.
PHARMACOKINETICS:
Penicillin V potassium is more resistant to inacrivation by gastric acid and is more completely absorbed than benzylpenicillin from the gastrointestinal tract following administration by mouth.Absorption is usually rapid. Peak plasma concentrations of 3 to 5 mcg/mfhave been observed 30 to 60 minutes after a dose of 500mg. The plasma half-life is about 30 - 60 minutes. About 80 % is reported to be protein bound it is metabolized in the liver. The unvhanged drug and metabolites are excreted rapidly in the urine.
INDICATIONS:
Penicillin V is used in the treatment or prophylaxis of infections caused by susceptible organisms, especially streptococci: Pharyngitis, tonsillitis. Rheumatic fever (primary and secondary prophylaxis), streptococcal skin infection. Pneumococcal infection prophylaxis and anthrax.
CONTRAINDICATIONS:
Patients with hypersensitivity to Penicillin's.
SIDE EFFECTS:
Penicillin V is well tolerated but may occasionally cause nausea, diarrhea, and hypersensitivity reactions, especially skin rashes anaphylactic reactions is rarely occur.
PRECAUTIONS:
It should be used with caution in patients with a history of allergy, especially to drug.
Renal and hematological status should be monitored during prolonged and high dose
therapy.
Care is necessary in patients with impaired renal function.
It should be given with caution during pregnancy and lactation
DRUG INTERACTIONS:
Guar gum may reduce the absorption of Penicillin V.
Probenecid prolongs the half-life of Penicillin V.
It should be used with care in patients taking nadolol and propranolol
Penicillin V should not be given with tetracycline or chloramphenicol concomitantly.
DOSAGE AND ADMINISTRATION:
Penicillin V is administered preferably at least 30 minutes before or 2 hours after food.
-Penicillin V 500: Adults: 1 2 tablets every 6 hours.
-Penicillin V 1000: Adults: 1 tablets every 6 hours.
-Penicillin V 400: children are given the following doses every 6 hours:
Up to 1 year: 62.5 mg, 1.25 ml (quarter small tea spoon) 1-5 years: 125 mg, 2.5 ml (half small tea spoon) 6-12 year: 250 mg, 5 ml (small tea spoon)
